Dean is transported back in time and is shocked when he lands in Lawrence, Kansas.
However, that shock turns to amazement after he runs into a young John Winchester (guest star Matthew Cohen) and Mary (guest star Amy Gumenick) who have just fallen in love. Dean isn't sure why he was sent back so he begins to enjoy the time with his parents and newfound grandfather (guest star Mitch Pileggi, “The X-Files”), who may be hiding a secret that is key to Sam (Jared Padalecki) and Dean's entire predicament.

Here's some more info...
Supernatural season 4 episode # 4 titled The Beginning.
It's meet the parents and see how they got to together as the story centers on John and Mary when they are in their twenties (not teens as originally made mention of).
John Winchester is just back from his tour in vietnam and ready to marry the love of his life- Mary. Mary loves John for who he is, a mechanic from a long line of mechanics and she wants to marry him too. The thing that stands in their way of happiness is Mary's father Samuel (played by the imposing and very tall Mitch Peliggi so now we know where Sam Winchester gets his name and his height).
Enter into the story, two mysterious young guys name 'Jeff' and 'Rick' who unbeknownst to Mary and John are their future sons Sam and Dean Winchester.
From what I gather from the information released I think that Jeff/Sam is apparently trying to warn Mary of coming future events and Rick/Jeff is bonding with John and having a few 'Back to the Future' homage style moments.
It sounds like it's a good episode and we may even found out that John has always had the Impala as his car.

I love finding out more about John and Mary, though the time traveling aspect has me a bit worried. But I'm confident Kripke will pull this off as well
